Whisperwake Isles opens after Aniimo's October 29 update, with three entry requirements: level 55, Grade I Trailblazer and completion of A Tale of Tide and Song. Reaching the level alone does not complete the unlock. The new story is part of the incoming update, so separate the progress you can make now from the story step that opens with it.
There is also a useful answer for players who will arrive later: the seasonal special Prismana Aniimo also appear in listed areas of the Breezy Plains. You do not need to unlock the new region to follow those announced Vein Abundance routes.

Treat the requirements as three checks on the same character. A level-55 character still needs the rank and story; reaching Grade I Trailblazer does not waive the level requirement.

The preview places Part II between the October 29 update and the December 10 update. Its start is tied to the update, rather than an announced midnight unlock. For now, the useful preparation is the level and rank work, followed by the named story when it becomes available.

Four areas give the new region different landscapes
Whisperwake Isles adds Crescent Bay, Rosy Mirelands, Wildwater Gorge and Cloudreach Falls. The announced views move from an open coastline through wooded wetlands and a river canyon to a landscape dominated by waterfalls:
The update also names three storylines: A Tale of Tide and Song, Secluded Seas, and Challenge of the Roaring Rapids. The first is the entry requirement; the other two are additional story content, rather than extra conditions listed for opening the region.
The Whisperwake Isles map provides a place to plan exploration. Keep the area names in view when reading the weekly event schedule: a route to Crescent Bay is different from one to Rosy Mirelands.
Separate isle Prismana forms from weekly specials
The region's new Prismana forms are Whirlbite, Gachapus, Jabster and Sugartini. Their preview points players toward nurturing the Whisperwake ecosystem. The broader update announces 32 new species and 51 new Aniimo; those are two different headline counts, not 51 separate species.
Vein Abundance is another layer. It highlights a special Prismana and an area pairing for a particular week. That schedule is the useful distinction when choosing a hunting destination: identify the named weekly target, then choose its listed isle or Breezy Plains location.
The Nurture guide explains the Branch, Bloom and regional-energy loop. The new calendar gives that loop a destination; it does not turn every creature shown in the update into the same acquisition route.

Each scheduled area pairing includes one location in Whisperwake Isles and one in Breezy Plains. In the table, Isles names the new-region destination and Plains names the alternative:

For players below the entry requirements, the Plains location in each pair is the important one. It supplies a concrete alternative to spending the week trying to reach level 55 before beginning the event hunt.
Example: a November 18 hunt below level 55
November 18 falls in the Jeburst week. A character who cannot enter Cloudreach Falls can use the listed Breezy Plains location, Russet Highlands, for that week's Vein Abundance route. The alternative changes the destination; it does not grant access to Whisperwake Isles or complete its story gate.
The opening October 30–November 1 row has a slash in the preview's special-creature column, so it does not name a special target. The final row is a Carnival period, rather than another single-creature week.
Four seasonal rewards use different acquisition systems
Some of Part II's creatures come through progression and events instead of the weekly Nurture hunt:

A Hypnyto goal therefore points toward the Handbook, while an Ekrim goal points toward seasonal achievements. Neither is the named target in the four weekly special-Prismana rows above. Hoptail and Jaguaraun also begin later, so their windows do not start on update day.

Choose the route that matches your actual goal
For new exploration, the route is level 55, Grade I Trailblazer, then the named story requirement. For a weekly special Prismana, the route is the dated area pairing, with a Breezy Plains option before you qualify for the isles. For Hypnyto, Ekrim, Hoptail or Jaguaraun, use the separate acquisition system in the reward table.
Those choices let you prepare without treating access to the new map as access to every new reward. Finish the missing entry checks for exploration, and put the specific creature's dates and acquisition method beside your collection goal.
